Determination of 2'-Fucosyllactose and Lacto-N-neotetraose in Infant Formula
A method for simultaneous determination of 2'-fucosyllactose(2'-FL)and lacto-N-neotetraose(LNnt)in infant formula by HPLC(high performance liquid chromatography)was established.The samples were extracted with water at 70 ℃ and derived from 2-o-aminobenzamide(2-AB)to obtain stable OS-2AB derivatives.The extracted solution was diluted with acetonitrile.The sample was detected by HPLC-fluorescence detector and quantified by external standard.In the concentration range of 10.0-200 μg/L,there was a good linear relationship between 2'-FL and LNnt,and the linear correlation coefficients were greater than or equal to 0.999.The limits of detection of 2'-FL and LNnt were 12.0 mg/100 g,and the limits of quantitation were 36.0 mg/100 g.The recoveries of 2'-FL and LNnt in blank infant formula samples with 3 concentration levels were investigated.The recoveries of 2'-FL and LNnT were in the range of 90%-105%,and the relative standard deviations were 1.29%-3.77%.The method has high sensitivity,good selectivity,accuracy,reliability and simple operation,and can be used for the detection of 2'-FL and LNnt in infant formula food.
