Optimization of extraction of polysaccharide from cuttlefish ink
Extraction of the cuttlefish ink polysaccharide was studied through single-factor experiment and orthogonal test.The ratio of material to liquid,pH,temperature and extracting time were studied by extraction rates of polysaccharides.Also activated carbon adsorption and high-speed centrifugation on the removal of the effect of Sepia melanin was compared.The results showed that activated carbon adsorption was better than high-speed centrifugation,the optimum conditions of extraction squid ink polysaccharide was:liquid ratio 1:2,pH5,temperature 40℃ and 4h for extracting, at this point the yield of polysaccharides was 1.228%, the content of polysaccharides was 78%After purified,protein removal rate was 90.52% ,the loss rate of polysaccharides was 12.45%.
