Study on the polysaccharide accumulation of Dendrobium officinale in rapid propagation
To obtain fast-breeding and high medicinal-value.D.officinale,polysaccharide content was identifying indicator.Taking PLB as a carrier,Nitrate/ammonium molecule ratio,sucrose and BA concentration were recognized the most important factors in seven medium compositions affecting PLBs polysaccharide content through the Plackett-Burman design.The steepest ascent experiment and further Box-Behnken design response surface methods were designed,optimal results of the presence of regression model were as follows:nitrate/ammonium molecule ratio,sucrose and BA concentration were 1.90∶1,29.1 g/L,1.65 mg/L.Under these conditions,the mass fraction of the polysaccharide increased 33.95% with a significant improvement compared with the value of 25.36% before optimization.The optimization effect was remarkable.
