Determination of galactooligosaccharide by high performance liquid chromatography
HPLC method was established to determine the galactooligosaccharide in dairy products and syrup. Glucose, galactose and lactose were simultaneously assayed on a NH2 column by using a refractive index detector and a mobile phase of acetonitrile-water. β-Galactosidase can hydrolyze GOS into galactose. Then GOS are calculated from concentrations of lactose and galactose. The relative standard deviation was 2.72%, 4.16%, 1.16%, 4.26% for glucose, galactose, lactose and galactooligosaccharide respectively. The method presented linear correlation coefficient between 0.9990 and 0.9995. The recovery ranged from 95% to 110%. This method was proved to be accurate and rapid.
