Optimization of Compound Enzyme Extraction Process and Antioxidant Activity of Epimedium koreanum Nakai Polysaccharides
Objective:To optimize the process of extracting polysaccharides from Epimedium by composite enzyme method and evaluate the antioxidant activity of Epimedium polysaccharides.Methods:Epimedium koreanum Nakai was used as the raw.material,and the polysaccharide extraction rate was used as the index.The polysaccharide of Epimedium was extracted by the synergistic effect of cellulase,pectinase,and papain.The extraction conditions of complex enzyme were optimized based on orthogonal test and response surface test.The antioxidant activity of Epimedium polysaccharide in vitro was evaluated by DPPH method and ABTS method.Results:The optimal enzyme extraction process was as follows:enzyme addition(pectinase 8000 U/g,cellulase 17600 U/g,papain 900 U/g),extraction time 2 h,pH 6.0,liquid to material ratio 53 mL/g,extraction temperature 50 ℃.Under these conditions,the yield of Epimedium polysaccharide was(16.95±0.15)%,3.86%higher than that of water extraction.Moreover,enzyme extracted polysaccharides have stronger in vitro antioxidant activity.Conclusion:The optimized process conditions of composite enzyme extraction are mild,the operation is simple,and the polysaccharide yield and antioxidant activity are higher than traditional water extraction methods.
