Optimization of Ultrasonic Assisted Deep Eutectic Solvents Extraction Process of Polyphenols from Mulberry Leaf and Its Antioxidant Activity
Using autumn mulberry leaf powder as raw material,the ultrasonic assisted deep eutectic solvents extraction method was applied to extract polyphenols.The optimal deep eutectic solvent composition was selected from among 20 types of deep eutectic solvents for extracting polyphenols from mulberry leaves.Based on single-factor experiments,the extraction process parameters were further optimized using response surface methodology,and the antioxidant capacity of mulberry leaf polyphenols in deep eutectic solvent extracts was analyzed by scavenging DPPH and ABTS+ free radicals.The results showed that the optimal extraction conditions for mulberry leaf polyphenols were as follows:choline chloride/fructose/alcohol molar ratio of1∶1∶2,water content of 45%,liquid-solid ratio of 40 mL/g,ultrasonic power of 360 W,ultrasonic temperature of 40 ℃,and an ultrasonic time of 40 min.Under these conditions,the amount of extracted polyphenols was 76.82 mg/g.In vitro antioxidant test results showed that the antioxidant ability of mulberry leaf polyphenols had a positive correlation with concentration and exhibited a significant scavenging effect on both DPPH free radicals and ABTS+ free radicals.
