双水相萃取地皮菜中藻蓝蛋白的研究

采用单因素实验和正交实验法对双水相萃取纯化地皮菜藻蓝蛋白技术进行优化.研究PEG1000/硫酸铵体系中PEG 1000含量、硫酸铵含量及离子强度对地皮菜藻蓝蛋白纯化的影响,以回收率为检验指标,确定了双水相萃取纯化地皮菜藻蓝蛋白的优化条件.结果表明,通过正交实验发现3种因素对地皮菜藻蓝蛋白回收率的影响大小依次为硫酸铵>氯化钾>PEG1000;并得到最优组合:双水相体系总质量为20 g,加入藻蓝蛋白粗提液5g时,PEG1000质量分数为14%、硫酸铵质量分数为15%、氯化钾质量分数0.5%,得到藻蓝蛋白纯度为0.302,回收率为96.43%.
Extraction of Phycocyanin from Nostoc commune by Aqueous Two-Phase System
The single factor experiment and orthogonal experiment were used to optimize the technology of aqueous two-phase extraction and purification of phycocyanin from Nostoc commune.The effects of PEG1000 content,ammonium sulfate content and ionic strength on the purification of phycocyanin from Nostoc commune were studied in PEG1000/ammonium sulfate system,The recovery rate was used as the test index to determine the optimal conditions of aqueous two-phase extraction and purification of phycocyanin from Nostoc commune.The results showed that the effects of three factors on the recovery rate of phycocyanin from Nostoc commune were in the order of ammonium sulfate>potassium chloride>PEG1000 in an orthogonal test.The optimal combination was obtained:with the total mass of 20 g aqueous two-phase system,join 5 g of phycocyanin crude extract,with PEG1000 mass fraction set at 14%,ammonium sulfate mass fraction at 15%,and potassium chloride mass fraction at 0.5%,the purity of phycocyanin was 0.302,and the recovery rate was 96.43%.
