Simultaneous Determination of 27 Free Nutrients in Rosa roxburghii Juice by Ultra-High Performance Liquid Chromatography-Tandem Mass Spectrometry
Objective:A method was established for simultaneous determination of 27 free nutrients in Rosa roxburghii juice by ultra-high performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)technology.Methods:Samples were extracted with 50:50(V∶V)acetonitrile/water,separated on an Agilent Poroshell 120 HILIC-Z column(2.1 mm×100 mm,2.7 μm)and with the gradient elution mobile phase of 20 mmol/L ammonium formate aqueous solution-20 mmol/L ammonium formate acetonitrile solution.The mass spectrometry detection was carried out by means of electrospray ionization in positive and negative mode with multiple reaction monitoring(MRM)method.Results:The developed method showed good linearity(R2≥0.995)for all the analytes within the test ranges,the limits of detection(LODs)ranged from 0.050 to 0.90 mg/kg,and the limits of quantification(LOQs)ranged from 0.20 mg/kg to 3.00 mg/kg.The spiked recoveries varied between 80.11%and 107.13%,and the relative standard deviations(RSDs)were 2.97%-9.82%.The application of the method to the analysis of the total amount of free amino acids in 4 batches of Rosa roxburghii juice samples range from 1405.66 mg/kg to 2061.28 mg/kg,the total amount of free vitamin B compounds were 6.98-10.56 mg/kg,and the average content of free catechin was 517.97 mg/kg.Conclusion:The method was simple,rapid,with good stability,and suitable for the simultaneous determination of 27 free nutrients in Rosa roxburghii juice.
