Inhibitory Activities of Portulaca oleracea L.Extracts onα-Amylase and α-Glycosidase
Objective:To further investigate the hypoglycemic activity and hypoglycemic mechanism of dried and fresh Portulaca oleracea L.(purslane).Methods:Ethanol extracts were obtained from dried purslane(DEP)and fresh purslane(FEP),after filtration and concentrate.Ethanol extract of dried and fresh purslane were extracted with n-hexane,ethylacetate and n-butanol,the DEP was further separated by different polar solvents and obtained four components:H-DEP,E-DEP,D-DEP,and W-DEP.the FEP was further separated by different polar solvents and obtained four components:H-FEP,E-FEP,D-FEP,and W-DFP.The inhibition activity on α-amylase and α-glucosidase were determined.Results:E-FEP showed the best inhibition effect for both enzymes,with IC50 values of(0.094±0.001)mg/mL and(33.63±0.42)μg/mL.The Lineweaver-Burk curve was plotted to determine the type of enzyme inhibition.The results showed that H-DEP,E-DEP,H-FEP and E-FEP were sensitive to α-amylase,which were the type of noncompetitive inhibition.DEP,FEP,D-DEP,W-DEP,D-FEP and W-FEP were competitive and non-competitive mixed inhibition.For α-glucosidases,the results showed that H-DEP,E-DEP,H-FEP and E-FEP were the type of noncompetitive inhibition.DEP,FEP,D-DEP,W-DEP,D-FEP and W-FEP were anti-competitive and non-competitive mixed inhibition type.Conclusion:Purslane has good hypoglycemic activity.The research results provide a theoretical basis for the comprehensive utilization of purslane resources,and are of great significance for the research and development of hypoglycemic foods and the advancement of hypoglycemic activity research.
