Optimization of Extraction Process and Antioxidant Activity of Lilium davidii var.Unicolor Salisb Polysaccharide
Using Lilium davidii var.Unicolor Salisb(Lanzhou lily)as raw materials,the process conditions for extracting polysaccharides from Lanzhou lily using enzymatic methods were explored,and the moisture absorption,moisturizing and antioxidant activities of polysaccharides were studied.On the basis of single factor experiments,Box Benhnken experiment was conducted with three factors and three levels,including enzyme dosage,enzyme hydrolysis time,and enzyme hydrolysis temperature.Data analysis was conducted using Design Expert software to obtain the optimal extraction conditions.The moisture absorption,moisturizing,and antioxidant activities of different intercepted fraction of lily polysaccharides obtained under the optimal extraction conditions by polysaccharide dialysis were studied.The results showed that the optimal extraction conditions for lily polysaccharides were enzyme addition of 4%,enzymatic hydrolysis time of 60 minutes,and enzymatic hydrolysis temperature of 55 ℃.Under these conditions,the polysaccharide yield was 25.91%.Through moisture absorption and moisturizing experiments,the best moisture absorption and moisturizing properties of lily polysaccharides were screened by intercepting molecular weight fragments of 3500-8000 u.After being placed under RH 43%,68%,and 93%conditions for 72 hours,the moisture absorption rates of 3500-8000 u molecular weight fragments were the highest,accounting for 62.67%,122.3%,and 119.2%,respectively.At RH 43%,the moisture retention rates of 3500-8000 u molecular weight fragments were the highest,accounting for 70.44%.In vitro antioxidant experiments showed that 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid)(ABTS)radical cation and hydrogen peroxide scavenging activities of 3500-8000 u fragments were higher than those of lily polysaccharide.The screening of molecular weight fragments intercepted by lily polysaccharides through moisture absorption and moisturizing tests provides a preliminary basis for the research of lily polysaccharides in cosmetics.
