Distribution of Puerarin Content in Fresh Pueraria thomsonii Benth and Simultaneous Extraction Technology of Starch and Puerarin
Puerarin is the main active ingredient of Pueraria thomsonii Benth,but in the process of producing pueraria starch,a large amount of puerarin is lost with the processing wastewater,resulting in resource waste.Simultaneous recovery of puerarin from wastewater during the extraction of pueraria starch is an important way to achieve high-value utilization of pueraria resources.The article used high-performance liquid chromatography to analyze the distribution pattern of puerarin content in different parts of pueraria and in the wastewater of pueraria starch extraction process.Single factor experiments and orthogonal experimental designs were used to optimize the simultaneous extraction process of pueraria starch and puerarin.The results showed that over 80%of puerarin was concentrated in pueraria bark,and the content of puerarin in the first starch extraction wastewater accounted for about 70%of the total wastewater.The optimal process for simultaneous extraction of starch and puerarin from fresh pueraria is to simultaneously add 2%D101 macroporous resin during the first extraction of starch,with an adsorption time of 90 minutes,an adsorption stirring speed of 150 r/min,a desorption ethanol concentration of 60%,a desorption agent volume of 30%wastewater,desorption twice,and a desorption time of 30 minutes each time.By using simultaneous extraction technology,the recovery rate of puerarin in the first washing wastewater during starch extraction can reach over 85%,and the extraction time can be effectively shortened to avoid the impact of microbial fermentation,physical and chemical reactions,etc.on the recovery efficiency of puerarin in the wastewater during the settling process of pueraria starch.This provides a scientific basis for the high-value utilization of pueraria resources.
