猫爪草多酚双水相提取工艺优化及抗氧化活性研究

通过优化猫爪草多酚的提取工艺及分析其抗氧化活性,以提高猫爪草的综合利用率.研究采用超声辅助PEG6000-柠檬酸钠双水相法提取猫爪草多酚,通过响应面法优化其提取工艺,并从多酚提取液对1,1-二苯基-2-三硝基苯肼(1,1-Diphenyl-2-picrylhydrazyl,DPPH)自由基清除率、2,2-联氨-双-3-乙基苯并噻唑啉-6-磅酸(2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid),ABTS)阳离子自由基清除率和铁离子还原能力3个方面分析其抗氧化活性.结果表明,猫爪草多酚的最佳提取工艺条件为:柠檬酸钠质量分数15%、PEG6000添加量1.6g、投料质量0.8 g、超声时间30 min,在此条件下,多酚提取量为0.408 mg/g,约是文献报道的1.6倍.该法得到的猫爪草多酚提取液对DPPH自由基和ABTS阳离子自由基均具有较好的清除能力,其IC50值分别为1.302 mg/L和0.6624 mg/L,且具有一定的还原能力.因此,超声辅助双水相法作为一种新型提取和分离方法,可高效、快速提取猫爪草中的多酚.猫爪草多酚具有良好的体外抗氧化活性,可进一步开发为天然抗氧化剂.
Optimization of Aqueous Two-Phase Extraction Process and Evaluation of Antioxidant Activity of Polyphenols from Ranunculi ternati Radix
In order to improve the comprehensive utilization rate of Ranunculi ternati Radix,the study optimized the extraction process of polyphenol and analyzed its antioxidant activity.The extraction process of polyphenols was optimized using the response surface methodology.The antioxidant activity of polyphenol extract was analyzed in terms of 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical clearance and 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid)(ABTS)radical cation clearance and ferric ion reducing antioxidant power(FRAP).The results showed that the optimum extraction conditions were determined as the content of sodium citrate was 15%,the dosage of PEG6000 was 1.6 g,the feeding mass was 0.8 g and the ultrasonic time was 30 min.Under these conditions,the yield of polyphenols was 0.408 mg/g,which was about 1.6 times of that reported in the literature.Ranunculi ternati Radix polyphenol extracts obtained by this method had good scavenging ability for DPPH radical and ABTS radical cation with IC50 values 1.302 mg/L and 0.6624 mg/L,respectively and had certain reducing ability.Therefore,as a new extraction and separation method,the ultrasonic-assisted aqueous two-phase method can extract polyphenols of Ranunculi ternati Radix efficiently and quickly.Moreover,the polyphenols of Ranunculi ternati Radix has good antioxidant activity in vitro and can be further developed as natural antioxidants.
