Simultaneous Analysis of Multiple Nucleosides/Nucleotides in 11 Kinds of Processed Food by Ultra-High-Performance Liquid Chromatography-Tandem Mass Spectrometry
Nucleotides and nucleosides are widely distributed in various types of food and condiments.A method using ultra-high-performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)was developed for the simultaneous detection of 11 nucleotide/nucleoside(adenosine,guanosine,cytidine,uridine,inosine,thymidine,adenosine monophosphate,guanosine monophosphate,cytidine monophosphate,uridine monophosphate,and inosine monophosphate)in processed foods.The results showed that the developed method had good linearity within the concentration range of the analyzed compounds.The linear range for the five nucleosides was from 0.080 μg/mL to 80.000 μg/mL,while for the six nucleotides,it was 0.002-2.000 μg/mL and 0.005-5.000 μg/mL,respectively,with the correlation coefficients above 0.9920,and each component was well separated.The limits of detection ranged from 0.293 to 9.186 ng/mL,and the limits of quantification ranged from 2.000 ng/mL to 80.000 ng/mL.The method exhibited good precision,repeatability,and stability,with average recovery rates ranging from 91.9%to 106.3%for dry sample,and average recovery rates ranging from 86.5%to 97.2%for wet sample.The method was applied to the quantitative analysis of nucleotides and nucleosides content in 11 commercially available food samples,and the results were in agreement with previously reported values,demonstrating the feasibility and reliability of the method.This developed UPLC-MS/MS method offers advantages over traditional high-performance liquid chromatography,including simple sample preparation,shorter analysis time,and lower cost.It provides a basic for simultaneous detection of nucleotide/nucleoside substances in processed food and serves as technical support for the development and quality testing of related products.
