摘要

L-缬氨酸是一种人体必需氨基酸,可应用于食品领域.L-缬氨酸主要通过微生物发酵法生产,而高产菌株是发酵工业的核心.为获得能大量积累L-缬氨酸的发酵菌株,以黄色短杆菌(Brevibacterium flavum)FP-01为诱变出发菌株,经常压室温等离子体、紫外线、硫酸二乙酯和亚硝基胍的多次诱变处理,通过选育氨基酸结构类似物抗性与氨基酸营养缺陷突变株,获得一株L-缬氨酸的高产突变菌株B.flavum FP-6-56,该突变菌株摇瓶发酵的L-缬氨酸产量达54.5 g/L.遗传稳定性试验结果表明,突变菌株B.flavum FP-6-56的遗传标记和L-缬氨酸产量具有较强的稳定性.通过诱变育种获得了 一株遗传稳定性高的L-缬氨酸高产突变菌株.

Abstract

L-valine is an essential amino acid.It is applied in food field.L-valine is mainly produced by microbial fermentation.High yielding strain is the foundation of the fermentation industry.Brevibacterium flavum FP-01 was mutated by atomspheric and room temperature plasma,ultra violet,diethyl sulfate,and nitrosoguanidine to breed mutants overproducing L-valine.A high yielding mutant strain,B.flavum FP-6-56,was obtained.It was resistant to amino acid structural analogues and was amino acid auxotrophic.54.5 g/L of L-valine was produced by the mutant strain in shaking flask.The results of genetic stability tests indicated that the genetic marker and L-valine production of B.flavum FP-6-56 were highly stable.A high-yield L-valine mutant strain with high genetic stability was obtained by mutation breeding.
