Optimization of Extraction Process by Response Surface Methodology and Antioxidant Activity of Total Flavonoids from Acorus tatarinowii Schott
In order to study the optimum ultrasonic extraction process of total flavonoids from Acorus tatarinowii Schott and evaluate its antioxidant activity,ultrasonic-assisted extraction was used to extract total flavonoids from these dried rhizomes.The extraction process of total flavonoids was optimized by single factor and response surface methodology.The antioxidant activities of total flavonoids were tested by 1,1-diphenyl-2-picrylhydrazyl(DPPH)assay and 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid)(ABTS)assay.The results showed that the optimum ultrasonic extraction parameters for total flavonoids from Acorus tatarinowii Schott were as follows:extraction time of 8 min,ethanol volume fraction of 85%,solid-liquid ratio 1∶40 g/mL,and the content of total flavonoids was 10.21 mg/g.The results of antioxidant capacity showed that total flavonoids from Acorus tatarinowii Schott displayed dose-dependent reducing power,positive linear correlation and strong abilities to scavenge DPPH radical and ABTS cation radical.And total flavonoids possessed free radical scavanging activity against ABTS cation radical(IC50=1.41 µg/mL)and DPPH radicals(IC50=7.75 μg/mL)in decreasing order as well as reducing power,the inhibiting abilities were far higher than Vitamin C as positive controls.These results demonstrate that total flavonoids from Acorus tatarinowii Schott have potent antioxidant activity,so it is worthy to be used as natural antioxidants.
