Construction the Method of Five Functional Components Simultaneous Determination and Fingerprint in Aquilaria sinensis Leaves
The high performance liquid chromatographic(HPLC)fingerprint of Aquilaria sinensis leaves and the simultaneous determination of its active ingredient content were established to provide scientific evidence for the quality control and exploitation of Aquilaria sinensis leaves.The extraction solvent and chromatographic conditions were optimized with 100%methanol extraction,Phenomenex-C18 column,methanol-pure water as mobile phase gradient elution,and the detection wavelength of 254 nm.The optimized method was systematically validated,and the linearity of the five constituents,including p-hydroxybenzoic acid,adenosine,arnica glycoside,lignoceroside,and tanshinone Ⅰ,was good(r≥0.9992),and the spiked recoveries were 96.53%-98.71%.The recoveries were high and the precision was good.The fingerprints of 32 batches of Aquilaria sinensis leaves were established,and the similarities were 0.889-0.998.The method intuitively reflected the differences in the chemical composition of Aquilaria sinensis leaves through the simultaneous determination of the contents of the five active ingredients,and provided a reference for the establishment of the quality evaluation standards for the industry of Aquilaria sinensis leaves industry in South China.
