Detection of Chlorpyrifos Residue in Cucumber Based on Aptamer-Suspension Chip
A method based on aptamer-suspension chip has been developed for the rapid detection of chlorpyrifos residues in cucumber.The aptamers of chlorpyrifos were covalently bound to carboxylated fluorescent microspheres to serve as detection probe,the 3'end complementary chains modified by biotin were used as competitive probe.The biotin-modified complementary chains could specifically bind streptavidin,R-phycoerythrin conjugates,resulting the complementary chain fluorescent and the content of chlorpyrifos was inversely proportional to the fluorescence intensity.Therefore,the content of chlorpyrifos in samples can be evaluated by fluorescence intensity.Under the optimized conditions,the developed method showed high sensitivity and specificity for chlorpyrifos.The detection concentration ranged from 0.039 ng/mL to 234.63 ng/mL,with a good linear connection(R2=0.98)and low limit of detection 0.014 ng/mL.The recovery of cucumber and the relative standard deviation were in ranges of 92.00%-105.30%and 1.76%-4.13%.The method exhibited high throughput,high sensitivity and good specificity,and had good application potential in the rapid detection of chlorpyrifos residues.
