摘要
通过单因素、Plackett-Burman试验及Box-Behnken试验优化培养基成分,进一步采用单因素及正交试验优化培养条件以提高植物乳植杆菌HYY-DB9活菌数.结果表明,植物乳植杆菌HYY-DB9的最佳培养基配方为海藻糖30g/L、磷酸氢二钾2g/L、乙酸钠3.59371 g/L、牛肉粉8g/L、酵母浸粉4g/L、细菌学蛋白胨25g/L、柠檬酸氢二铵4g/L、硫酸镁0.5 g/L、硫酸锰0.04g/L、吐温80 2.5 g/L;最佳培养条件为培养温度32 ℃、初始pH5.9、接种量2%.此时植物乳植杆菌HYY-DB9活菌数达到最大值6.9×109 CFU/mL,相较于优化前提高了4.6倍.该结果可为后续发酵制剂的制备及商业化应用提供参考.
Abstract
The medium composition was optimized by one-way,Plackett-Burman test and Box-Behnken test,and further one-way and orthogonal tests were used to optimize the culture conditions to increase the viable count of Lactiplantibacillus plantarum HYY-DB9.The results show that the optimal medium formulation of Lactiplantibacillus plantarum HYY-DB9 is as follows:trehalose 30 g/L,dipotassium hydrogen phosphate 2 g/L,sodium acetate 3.59371 g/L,beef powder 8 g/L,yeast extract 4 g/L,bacteriological peptone 25 g/L,diammonium hydrogen citrate 4 g/L,magnesium sulphate 0.5 g/L,manganese sulphate 0.04 g/L,Tween 80 2.5 g/L.The optimal culture conditions are:fermentation temperature 32 ℃,initial pH 5.9,and inoculum quantity 2%.At this time,the viable bacterial count of Lactiplantibacillus plantarum HYY-DB9 reached the maximum value of 6.9×109 CFU/mL,which was 4.6-fold higher compared to before optimisation,and provided a reference for the preparation and commercial application of the subsequent fermentation preparation.
