Optimization of Ultrasonic and Enzymatic Extraction Process of Polysaccharides from Nostoc sphaeroides Küte and Its Anti-Fatigue Activity in Wine Body
The extraction rate of polysaccharides from Nostoc sphaeroides Küte was used as the index,and on the basis of single factor experiments,the process conditions for extracting polysaccharides from Nostoc sphaeroides Küte by ultrasound combined with enzyme method were optimized through response surface methodology.The crude polysaccharides of Nostoc sphaeroides Küte was purified to obtain alcohol soluble components and the anti-fatigue effects of each purified polysaccharide component were investigated in wine body,respectively.Finally,the molecular weight range of the obtained components with anti-fatigue activity was determined by high-performance gel exclusion chromatography.The results showed that the optimal extraction rate of polysaccharides from Nostoc sphaeroides Küte was(38.57±0.47)%when the addition amount of composite enzyme was 2.0%,enzymatic hydrolysis pH was 4.9,enzymatic hydrolysis time was 2.0 h,enzymatic hydrolysis temperature was 50 ℃,ultrasonic power was 163 W,and solid-liquid ratio was 1∶200 g/mL.The crude polysaccharide from Nostoc sphaeroides Küte was separated and purified by DEAE Seplife A25 glucan gel chromatography column,and two polysaccharide components were obtained:GXM-1 and GXM-2,respectively.The molecular weight distribution ranges were concentrated in(7×105-60×105)u and(9×103-6×104)u,respectively.The results of the anti-fatigue study showed that both GXM-1 and GXM-2 at a medium dose had a strong anti-fatigue activity.This study can provide data support for the development and utilization of functional foods containing the polysaccharides from Nostoc sphaeroides Küte.
