Optimization of Ultrasound-Assisted Extraction of Alkaloids from Amomum tsaoko by Response Surface Methodology
Objective:To optimize the quantitative determination method and extraction conditions of alkaloids from Amomum tsaoko.Method:With berberine hydrochloride as the reference substance,the detection conditions of acid dye colorimetric method for quantitative determination of alkaloids in A.tsaoko were optimized by single factor experiment design;The effects of ethanol concentration,solid-liquid ratio,ultrasonic power,ultrasonic time and ultrasonic temperature on the extraction rate of alkaloids from A.tsaoko were investigated with the total alkaloids content as the index.On this basis,the extraction conditions were optimized by response surface design;Then the effect of pH value of acid extraction and alkali precipitation on the yield of alkaloids extracted from A.tsaoko by organic solvent method was explored.Results:The detection methods of alkaloids in A.tsaoko were as follows:1 mL of sample solution,2 mL of phosphate buffer solution with pH value of 2.0,300 µL of Bromocresol green,1 mL of chloroform extraction solution and 0.5 g of anhydrous sodium sulfate were mixed and stood for 30 min,and the chloroform layer was detected at 410 nm;The optimum extraction conditions of alkaloids from A.tsaoko were as follows:ethanol concentration was 65%,the ratio of material to solvent was 1∶12(g/mL),the ultrasonic power was 160 W,the pH value of acidic solution was 4.0,and the pH value of alkaline solution was 10.0.Under these conditions,the extraction rate of alkaloids from A.tsaoko was 0.818%.Conclusion:The optimized method for the determination of alkaloids in A.tsaoko has the advantages of high precision,the optimized extration technology has the advantages of simple operation,reliable process parameters and good extraction effect,which provides an important basis for the further study of alkaloids in A.tsaoko.
