高效阴离子交换色谱-脉冲安培法检测乳制品中7种稀有糖

文章建立了高效阴离子交换色谱-脉冲安培法测定乳制品中7种稀有糖的分析方法.样品经超声提取、沉淀蛋白后,采用CarboPac PA20(3 mm×150 mm)色谱柱分离,在流速为0.40 mL/min,柱温为25 ℃的梯度洗脱条件下进行分析,外标法定量.7种稀有糖(海藻糖、岩藻糖、阿拉伯糖、塔格糖、山梨糖、阿洛酮糖和异麦芽酮糖)在0.2~50.0 mg/L浓度范围内线性相关系数良好(r≥0.999),方法的定量限为0.05~0.28 mg/100 g.7种稀有糖的回收率为86.9%~106.0%,相对标准偏差为0.29%~3.12%.该方法在15 min内即可完成7种稀有糖的定量分析,操作简便、分析速度快、灵敏度高,可以应用于乳制品中多种稀有糖含量的测定.
Determination of Seven Rare Sugars in Dairy Products by High Performance Anion Exchange Chromatography with Pulsed Amperometry
An analytical method for the determination of seven rare sugars in dairy products by high performance anion exchange chromatography with pulsed amperometry was established.After ultrasonic extraction and protein precipitation,the samples were separated by CarboPac PA20(3 mm×150 mm)column and analyzed under gradient elution conditions of 0.40 mL/min flow rate and 25 ℃ column temperature.Quantification was performed using the external standard method.The seven rare sugars(trehalose,fucose,arabinose,tagatose,sorbose,aloketose,and isomaltulose)showed good linearity in the concentration range of 0.2 mg/L to 50.0 mg/L,with correlation coefficients r≥0.999.The limits of quantification ranged from 0.05 mg/100g to 0.28 mg/100g.The recoveries of the seven rare sugars ranged from 86.9%to 106.0%,with relative standard deviations from 0.29%to 3.12%.The method allows for the quantitative analysis of the seven rare sugars within 15 minutes,offering simplicity,rapid analysis,high sensitivity,and applicability for measuring multiple rare sugars in dairy products.
