实时荧光PCR法检测鸭血制品中其他动物源性成分

目的:为对市售鸭血制品进行掺假检测,文章建立了实时荧光聚合酶链式反应法(Polymerase chain reaction,PCR)检测鸭血中鸡、鸭、鹅、猪、牛、羊等动物源性成分的定性检验方法.方法:考察了3种不同的DNA提取方式对检测结果的影响,确定了方法的检出限,并对抽取的51份市售鸭血样品进行检测.结果:烘干称质量提取的DNA浓度及扩增效率最高,该方法灵敏度可达到1%.51份实际样本中共发现8批样本存在掺假行为.结论:该方法可实现对鸭血中其他动物源性成分的定性检测,为执法监管部门提供有力的技术保障.
Qualitative Detection Methods of Animal Derived Ingredients in Duck Blood by Real-Time Fluorescence PCR
Objective:In order to detect the adulteration of animal derived ingredients in duck blood products,a qualitative detection method was developed in this paper for real-time fluorescence polymerase chain reaction(PCR)of animal derived components such as chicken,duck,goose,pig,cow,and sheep in duck blood.Method:The method investigated the impact of three different DNA extraction methods on the results,explored the detection limit of the method,and tested 51 duck blood samples.Results:The DNA concentration and amplification efficiency extracted by drying and weighing were the highest,and the sensitivity of the method could reach 1%.In actual samples detection,8 out of 51 duck samples were found to have other animal derived ingredients.Conclusion:This method realize the qualitative detection of other animal derived components in duck blood,providing strong technical support for law enforcement and regulatory authorities.
