Extraction,Purification and Structural Analysis of Brassica rapa L.Polysaccharides
Objective:The extraction process of Brassica rapa L.polysaccharides(BRAP)was optimized with the assistance of ultrasonication,followed by purification and structural analysis of the polysaccharides.This study not only offers valuable insights into future research on BRAP and their biological activities but also lays a solid foundation for further exploration in this field.Methods:The ultrasonic-assisted extraction process was optimized by a combination of single factor test and response surface test,BRAP were separated and purified by DEAE cellulose-52 column and Sephadex G-100 gel chromatography column,and the structure of BRAP was identified by spectroscopy,chromatography and electron microscopy.Results:The optimal extraction process of BRAP was 175 W with ultrasonic power,60 ℃,solid-liquid ratio 1:30 g/mL,and ultrasonic time of 60 min,and the highest yield of BRAP was 41.12%.Three BRAP components were isolated and purified,and the highest component crude polysaccharide BRAP-1 was further separated and purified to obtain uniform BRAP1-1.BRAP1-1 was determined by liquid chromatography to be composed of fructose,rhamnose,arabinose,galactose and galacturonic acid,with a molar ratio of 0.81∶4.30∶3.61∶1.69∶89.59.Conclusion:The ultrasonic-assisted extraction process of BRAP is greatly improved,and the spectral scanning results show that BRAP1-1 has the characteristic absorption peak of polysaccharides.
