Ameliorative Effect of Hovenia dulcis Peduncle Polysaccharides on Alcohol-Induced Ferroptosis in sh-sy5y Cells
In order to investigate the ameliorative effect of Hovenia dulcis fruit peduncle polysaccharides(HDPs)on alcohol-induced ferroptosis in sh-sy5y cells,sh-sy5y cells were treated with different concentrations of alcohol and HDPs.The contents of reactive oxygen species(ROS),malondialdehyde(MDA),4-hydroxynonenal(4-HNE),glutathione(GSH),Fe2+,and glutathione peroxidase 4(GPX-4)activity were measured to clarify the induction of ferroptosis by alcohol in sh-sy5y cells and the ameliorative effect of HDPs.At the same time,the key indicators of the Warburg effect,pyruvate dehydrogenase kinase isozyme 1(PDK-1),lactate dehydrogenase A(LDHA),and lactate expression were measured to explore the potential mechanism of alcohol-induced ferroptosis in sh-sy5y cells and the ameliorative effect of HDPs.The results showed that after exposure to alcohol,the contents of ROS,4-HNE,Fe2+,and MDA in sh-sy5y cells increased;the cell viability,GSH level,and GPX-4 activity decreased;at the same time,the expression levels of PDK-1,LDHA,and lactate increased,while HDPs could significantly inhibit these changes.The results indicated that alcohol exposure could induce ferroptosis in sh-sy5y cells and HDPs could ameliorate the ferroptosis.The ameliorative effect related to down-regulating the Warburg effect during ferroptosis.The research results provide a reference for clarifying the ameliorative effect of HDPs on alcohol-induced nerve injury,which is significance for promoting the development and utilization of Hovenia dulcis resources.
