Dynamic Ultra high Pressure Micro Jet Homogenization Assisted Ethanol Extraction of Rooibos Flavonoid and Its Regulation of Blood Glucose
Using rooibos as raw material,its flavonoids were extracted by dynamic ultra high pressure micro jet homogenization assisted ethanol method.The extraction process was optimized by response surface methodology,and its hypoglycemic effects were studied.The results showed that the optimal extraction process conditions were:liquid-to-solid ratio was 29∶1(mL/g),micro jet pressure was 147 MPa,and extraction time was 63 minutes.Under these conditions,the yield of rooibos flavonoids was 2.86%.The results of the hypoglycemic effect test in mice showed that rooibos flavonoids could reduce body weight and blood glucose levels,improve glucose tolerance,reduce total cholesterol(TC),triglyceride(TG),and low-density lipoprotein cholesterol(LDL-C)in mouse serum,increase high-density lipoprotein cholesterol(HDL-C),reduce malondialdehyde(MDA)content,increase the activity of catalase(CAT),glutathione(GSH),and superoxide dismutase(SOD)in mice.This indicates that rooibos flavonoids have the effect of regulating blood sugar,as well as regulating blood lipids and antioxidant activity.This experiment can provide a reference for the extraction and product application development of rooibos flavonoid.
