Determination of Nucleotides and Nucleosides in Infant Formulae and Special Medical Formulas by Liquid Chromatography-Tandem Mass Spectrometry
A liquid chromatography-tandem mass spectrometry(LC-MS/MS)method was developed and optimized for the determination of five nucleotides,including uridine monophosphate(UMP),adenosine monophosphate(AMP),cytidine monophosphate(CMP),inosine monophosphate(IMP),and guanosine monophosphate(GMP)and five nucleosides,including uridine(U),adenosine(A),cytidine(C),inosine(I)and guanosine(G)in infant formulae and special medical foods.The samples were dissolved in water,and the proteins were precipitated by 50%acetic acid solution,and filtered.Waters HSS T3 column(2.1 mm×100 mm,1.8 μm)was used as the analytical column,and the gradient elution was carried out with the mobile phases of 50 mmol/L ammonium acetate(pH5.0)aqueous solution and methanol.The mass spectra were detected by electron spray ionization positive ion mode in selected reaction monitoring mode(SRM)and quantified by external standard method.The concentrations of the standard curves were all in the range of 10-600 ng/mL,and the correlation coefficients(r)were in the range of 0.9992-0.9998.The average spiked recoveries of the six parallel determinations of nucleotides and nucleosides were in the range of 95.1%-112%,and the relative standard deviations(RSDs)were in the range of 0.43%-2.86%.The limits of detection(LODs)were in the range of 0.1-0.3 mg/kg,and the limits of quantification(LOQs)were in the range of 0.2-1.0 mg/kg.The method is accurate,convenient,fast and sensitive,and can be used for the quantitative analysis of five nucleotides and five nucleosides in infant formulae and special medical foods.
