Purification of Glycoprotein from Polygonatum odoratum and Its Antioxidant Activity in vitro
Crude glycoproteins (PDGs) were obtained from Polygonatum odoratum through phosphate buffer extraction.PDGs were further purified by Sephadex G-100 gel filtration and concanavalin A affinity chromatography,and PDG1 and PDG3 were acquired.The molecular weights and antioxidant activity of these two fractions were determined.The results showed that PDG1 and PDG3 had a molecular weight of 186 and 28.3 kD,respectively,and PDG,PDG1 and PDG3 had reducing power,strong abilities to scavenge hydroxyl radical and 1,1-diphenyl-2-picrylhydrazy (DPPH) radical and inhibitory effects against lipid peroxidation,showing a dose-effect relationship in the range of 1-10 mg/mL.Generally,PDG1 and PDG3 exhibited better antioxidant activity than PDGs.
