A method for determination of aflatoxin B1 in food by using thioflavine T-based nucleic acid probe cascade immunofluorescence assay
To develop an accurate,sensitive,and specific method for the detection of aflatoxins B,(AFB,)in complex food matrix,antibody modified magnetic nano-microspheres(MMPMs-AFB,antibody)was used as the capture probes,and antigen/GOx modified nano-microspheres(AFB,-OVA-PPs-GOx)was used as the signal probes,combining with Fenton reaction-sulfur T(ThT)fluorescent nu-cleic acid probe system,a"turn on"type AFB1 magnetic separation fluorescence immunoassay method was developed.The detection range of AFB1 by this method was 0.05-50 ng/mL and the detection limit was 0.027 ng/mL.The concentration of the spiked sample was 1,5,20,200 ng/g(ng/mL),the recovery of AFB,in spiked foodstuffs ranged from 81.6%to 112.1%,with the coefficient of variation of 1.2%-15.8%.In quality control sample of corn flour,the method was successfully applied to detect AFB1.This method has high sensi-tivity,good specificity and strong anti-matrix interference ability,and can be used for accurate quantitative detection of AFB,in food.
