水酶法提取枸杞籽油工艺优化

用水酶法提取枸杞籽油,先通过单因素试验选取影响因素和水平,再采用二次正交旋转组合设计方法研究液料比、酶添加量、酶解时间对枸杞籽油得率的影响.结果表明,各因素对枸杞籽油得率影响大小依次为液料比>酶添加量>酶解时间;频率分析法得到枸杞籽油最优的提取工艺为液料比3.6∶1(V∶m),酶添加量3.0%(以原料量计),酶解时间3h,该条件下3次的枸杞籽油平均验证得率为87.6%.
Optimization of aqueous enzymatic method extraction of lycium seed oil by quadratic orthogonal rotation design
The extraction of lycium seed oil by aqueous enzymatic method was adopted.The experimental values were firstly selected by single-factor test and a quadratic orthogonal rotation combination design was chosen based on the results of single factor test in order to study the effects of three main technological conditions,including ratio of soild to solvent,dosage of Viscozyme L and enzymolysis time on yield of oil from lycium seed.Results showed that the influence of investigated facors on yield of lycium seed oil followed the decreasing order:ratio of soild to solvent>dosage of Viscozyme L> enzymolysis time and a maximum yield of 87.6% was achieved under the optimal extraction:ratio of soild to solvent 3.6 ∶ 1(V ∶ m),dosage of Viscozyme L 3.0% and enzymolysis time 3 h.
