富硒平菇多糖提取纯化及抗氧化活性研究

Extraction and Purification of Polysaccharide from Selenium Contained Pleurotus ostreatus and Antioxidant Activity Assay

从富硒平菇中提取粗多糖Se-POP,进一步分离纯化得到Se-POP-1,Se-POP-2,Se-POP-33个组分,3个组分中的总糖、糖醛酸、硫酸基的含量都较Se-POP有所提高,而硒含量基本不变,其中Se-POP-1中的各成分含量高于另外两种组分.不同的硒多糖组分对DPPH等自由基的清除率都随着浓度的增加而增大,同浓度下Se-POP-1对自由基的清除率显著高于Se-POP-2和Se-POP-3(P<0.05),6 mg/mL时Se-POP-1对DPPH自由基、ABTS+自由基、羟自由基和超氧阴离子自由基的清除率分别为82.16 %、56.18 %、76.16 %、79.25 %.
Crude selenium contained polysaccharide(Se-POP)was extracted from Pleurotus ostreatus fortified with sodium selenite,three purified fractions of Se-POP-1,Se-POP-2,Se-POP-3 were obtained by passing through DEAE-cellulose and Sephadex G-100 chromatographic column. The contents of total sugar, uronic acid and sulfuric acid in the three components were higher than that of Se-POP,while the content of selenium was almost not changed,and the sugar,uronic acid,sulfuric acid and selenium contents in Se-POP-1 were higher than that of Se-POP-2 and Se-POP-3.The scavenge rate for DPPH free radical and other free radicals was increased with the increasing concentration of Se-POP fractions, and clearance rate for Se-POP-1 was significantly higher than Se-POP-2 and Se-POP-3(P<0.05),the scavenge effect for Se-POP-1 with the concentration of 6 mg/mL on DPPH free radical, ABTS+radical, hydroxyl radical and superoxide anion free radical were 82.16 %,56.18 %,76.16 % and 79.25 %,respectively.
