过表达丙酮酸脱氢酶对高产柠檬酸菌株的影响

为研究丙酮酸脱氢酶对黑曲霉柠檬酸产量的影响,该文构建含有不同启动子的丙酮酸脱氢酶过表达质粒p14(pGAS)、p16(pGAP)并获得产酸性能好且遗传稳定的过表达菌株p14-45 和p16-5.30 L发酵罐发酵结果表明,突变菌株p14-45 和p16-5 在发酵结束后相对于出发菌株CGMCC 10142 的产酸量分别提高 5.17%、4.01%.
Effect of Overexpression of Pyruvate Dehydrogenase on Citric Acid-Producing Strains
In order to investigate the effect of pyruvate dehydrogenase on citric acid production in Aspergillus niger,overexpression plasmids of pyruvate dehydrogenase p14(pGAS)and p16(pGAP)with different pro-moters were constructed,and overexpression strains p14-45 and p16-5 with good acid-producing performance and genetic stability were obtained.The fermentation results in the 30 L fermenter showed that the acid produc-tion of mutant strains p14-45 and p16-5 increased by 5.17%and 4.01%,respectively,compared with that of the original strain CGMCC 10142 at the end of fermentation.
