为提高细菌胞外多糖(exopolysaccharides,EPS)的工业生产效率,该研究通过单因素试验优化一株波茨坦短芽孢杆菌S8 的产糖培养条件,进一步采用Plackett-Burman设计和最陡爬坡试验对最佳产糖培养条件进行响应面优化,并将菌株所产EPS分别与不同疏水底物混合,以吐温 80 作阳性对照,对其乳化性进行探究.结果表明:优化的最佳产糖条件为接种量 4.56%、培养时间 39.9 h、碳源添加量 30.04 g/L;在此条件下,EPS 产量可达到(107.81±1.47)mg/L;乳化性探究试验结果表明波茨坦短芽孢杆菌S8 产生的EPS对疏水性底物具有较好的乳化作用,12 h和24h的乳化效果明显优于吐温 80.
Culture Condition Optimization of a Brevibacillus borstelensis Strain for Producing Exopolysaccharides and Analysis of Exopolysaccharide Emulsifying Performance
The culture conditions of Brevibacillus borstelensis S8 for producing exopolysaccharides(EPS)were explored by single-factor experiments to improve the production efficiency of EPS.The culture conditions for producing EPS were optimized by Plackett-Burman design and steepest ascent experiments.The EPS produced by strain S8 was mixed with different hydrophobic substrates to investigate its emulsifying performance,with Tween 80 as a positive control.The results showed that the optimal culture conditions of the strain for EPS pro-duction were the inoculation amount of 4.56%,carbon source addition at 30.04 g/L,and culture for 39.9 h.Un-der these conditions,the EPS yield reached(107.81±1.47)mg/L.The EPS produced by B.borstelensis S8 had good emulsifying performance on hydrophobic substrates,with the emulsifying performance better than that of Tween 80 at the time points of 12 h and 24 h.
