HPLC Analysis of Effects of Different Extraction Methods on the Content of Three Components in Inonotus obliquus
A high performance liquid chromatography(HPLC)method was established to determine the con-tent of protocatechuic acid,protocatechuic aldehyde,and osmundacetone in the crude extract of Inonotus obliquus).The effects of four different extraction methods,i.e.,ultrasonic extraction,reflux extraction,microwave-assisted extraction,and enzyme-assisted extraction,on the content of active ingredients in the crude extract of I.obliquus were compared and evaluated.The results indicated that the assay had high precision,with relative standard deviations(RSD)of 0.14%,0.06%,and 0.20%for the three compounds.It also showed good reproducibility,with RSD values of 1.67%,1.98%,and 2.22%,respectively,and good stability over 24 h,with RSD values of 0.20%,0.61%,and 0.25%,respectively.A comparison of the effects of different extraction methods on the three compounds revealed that the target compounds obtained by ultrasound-assisted extraction gave higher yields and were suitable for the extraction of the main active components in I.obliquus.The HPLC method established in this study for determining the content of protocatechuic acid,protocatechuic aldehyde,and osmundacetone in the crude extract of I.obliquus produced accurate results.
