造血干细胞衰老分子机制研究进展

造血干细胞可以满足机体对各类血细胞的需要,随着年龄的增长,造血干细胞逐渐衰老,其功能减退、数量变化,生物体衰老起源于干细胞衰老,随着人口老龄化,衰老相关疾病日益受到关注,本研究通过细胞内在与外在因素2个方面讨论造血干细胞分子衰老机制.以"造血干细胞""衰老"为关键词,检索2018-01-01-2023-07-31中国知网及PubMed相关文献.纳入标准:造血干细胞衰老相关通路的研究;造血干细胞衰老和DNA损伤的研究;造血干细胞衰老和代谢的研究;造血干细胞衰老和表观遗传学的研究;造血干细胞衰老和环境的研究.排除标准:年份较远、质量较差的文献.最终纳入分析文献52篇.结果表明,造血干细胞衰老的分子机制涉及细胞内外因素,其中DNA损伤、代谢、表观遗传学、环境等因素参与造血干细胞衰老过程.进一步研究造血干细胞衰老相关分子机制,有望发现更多有效的延缓甚至逆转造血干细胞衰老过程的新靶点.
Research progress on molecular mechanism of hematopoietic stem cellular aging
Hematopoietic stem cells can meet the needs of the body for various types of blood cells.With age,hematopoiet-ic stem cells gradually age,their function decreases,and their quantity changes.Biological aging originates from stem cell aging.With the aging of the population,age-related diseases are increasingly receiving attention.This study discusses the molecular aging mechanism of hematopoietic stem cells from both internal and external factors of cells.Using the key-words"hematopoietic stem cells"and"aging",we searched for relevant literature on CNKI and PubMed from January 1,2018 to July 31,2023.Inclusion criteria:Research on senescence-related pathways of hematopoietic stem cells;Research on aging and DNA damage of hematopoietic stem cells;Research on aging and metabolism of hematopoietic stem cells;Research on aging and epigenetics of hematopoietic stem cells;Research on aging and environment of hematopoietic stem cells.Exclusion criteria:Documents that are older and of poorer quality.Finally,52 articles were included for analysis.The results indicate that the molecular mechanism of hematopoietic stem cell aging involves both intracellular and extracel-lular factors,among which factors such as DNA damage,metabolism,epigenetics,and environment are involved in the aging process of hematopoietic stem cells.Further research on the aging related mechanisms of hematopoietic stem cells is expected to discover more effective new targets for delaying or even reversing the aging process of hematopoietic stem cells.
