Expression of YES-associated protein 1 and their relationship with survival in breast cancer tissue
Objective T he function of transcriptional co-activator YES-associated protein 1(YAP1)was controversial in breast cancer.This study aimed to investigate the expression and function of YAP1 in invasive breast cancer tissues.Methods Clinical pathological features and survival data of 573 patients with invasive breast cancer who visited Qingdao University Affiliated Taian Central Hospital from January 1,2011,to January 1,2013,were collected.Immunohisto-chemistry was used to detect the expression of YAP1 in breast cancer tissues.The relationship between YAP1 expression and various clinicopathological features was analyzed using the chi-square test and Spearman's correlation test.Kaplan-Meier univariate analysis and Cox multivariate regression analysis were used to assess the impact of YAP1 expression on the survival of breast cancer patients with different molecular subtypes.Results The study found that YAP1 was mainly localized in the cytoplasm and nucleus.YAP1 expression differed in different lymph node status(x2=7.876,P=0.049),TNM stage(x2=8.581,P=0.014),estrogen receptor(x2=6.201,P=0.013),progesterone receptor(x2=4.988,P=0.026),human epidermal growth factor receptor 2(x2=4.770,P=0.029),Ki-67(x2=6.103,P=0.013),and molecular subtypes(x2=17.472,P=0.002).YAP1 expression was positively correlated with tumor size(ρ=0.111,P=0.008),lymph node status(ρ=0.084,P=0.046),Ki-67 index(ρ=0.103,P=0.013),and TNM stage(ρ=0.122,P=0.003),but negatively correlated with estrogen receptor(ρ=-0.104,P=0.013),progesterone receptor(ρ=-0.093,P=0.026),human epidermal growth factor receptor 2(ρ=-0.091,P=0.029),and molecular subtypes(ρ=-0.131,P=0.002).Kaplan-Meier univariate analysis showed that tumor size,histological grade,lymph node sta-tus,estrogen receptor,progesterone receptor,Ki-67 positive index,TNM stage,molecular subtypes,and YAP1 expres-sion were important factors affecting disease-free survival(DFS)and overall survival(OS)in the overall population of breast cancer patients in this study,P<0.05.In multivariate Cox regression analysis,histological grade,lymph node status,progesterone receptor,and molecular subtypes were significant prognostic factors for DFS(P<0.05),while his-tological grade,molecular subtypes,and YAP1 expression were significant prognostic factors for OS,P<0.05.Specific analysis of different molecular subtypes showed that in Luminal A subtype,the impact of YAP1 expression on DFS was statistically significant(P=0.049),while the impact on OS was not statistically significant.In the triple-negative sub-type,YAP1 had a statistically significant impact on DFS(P=0.022)and OS(P=0.025)of patients.Conclusion YAP1 acts as an oncogene in invasive breast cancer and may be one of the indicators for predicting the prognosis of invasive breast cancer,especially triple-negative subtype.
