Study on the apoptosis induction of human ovarian cancer SK-OV-3 cells by activating NF-κB and inhibiting AMPK pathway with extracts from polygonum chinense
Objective To investigate the effect of polygonum chinense extract on apoptosis of human ovarian cancer SK-OV-3 cells and its possible molecular mechanism.Methods The active ingredients were extracted with ethyl acetate,and the effects of the extract on the proliferation of human ovarian cancer SK-OV-3 cells were detected by MTT,the cell cycle and apoptosis were detected by flow cytometry,and the Nuclear factor kappa B(NF-κB)and AMP-activated protein kinase(AMPK)pathway-related proteins were detected by protein blotting.Results Ethyl acetate extract of polygonum chinense could inhibit the proliferation of human ovarian cancer SK-OV-3 cells,and the inhibition rate increased with treatment concentration and time.When the concentration of Polygonum chinense was 10 mg/L,the mean inhibition rates of ovarian cancer cell proliferation at 24,48 and 72 hours were 3.37%,4.21%and 5.51%;when the concentration was 100 mg/L,the average inhibition rates of ovarian cancer cell proliferation at 24,48 and 72 hours were 38.86%,45.86%and 54.54%;when the concentration was 1 000 mg/L,the average inhibition rates at 24,48 and 72 hours were 65.39%,73.76%and 84.97%,with all P<0.01.The P values for apoptosis rates detected by flow cytometry 24 h after treat-ment with 0,10,20,50,100,and 1,000 mg/L of polygonum chinense ethyl acetate extract were 0.977,0.013,0.008,0.003,and 0.012,respectively,indicating that the apoptosis rate of SK-OV-3 cells significantly increased when treated with more than 20 mg/L of polygonum chinense ethyl acetate extract.Western blot results showed that the expression level of NF-κB p65 protein in SK-OV-3 cells increased with the increase of ethyl acetate extract concentration,while the expression level of AMPK protein decreased.Conclusions The ethyl acetate extract of polygonum chinense may induce apoptosis in human ovarian cancer SK-OV-3 cells by activating the NF-κB pathway and inhibiting the expression of AMPK protein.The toxic effects of ethyl acetate extract of Polygonum chinense on human ovarian cancer SK-OV-3 cells were dose-dependent and time-dependent.
