Expression of miR-136-5p and FAM83H in triple negative breast cancer and their correlation with clinicopathological characteristics and prognosis
Objective To investigate the expression of microRNA-136-5p(miR-136-5p)and family with sequence similarity 83 member H(FAM83H)in triple negative breast cancer(TNBC)tissues and their correlation with clinicopathological characteristics and prognosis.Methods A total of 172 patients with TNBC who underwent surgical treatment at the First Affiliated Hospital of Henan University from January 30,2015 to June 30,2018 were selected as the study subjects.Re-al-time fluorescence quantitative polymerase chain reaction(RTFQ-PCR)was applied to detect the expression of miR-136-5p and FAM83H in cancerous and paracancerous tissues.Pearson method was applied to analyze the correlation between miR-136-5p and FAM83H expression levels in cancer tissue of TNBC patients.Kaplan-Meier survival curve was applied to analyze the relationship between the expression levels of miR-136-5p and FAM83H in cancer tissue of TNBC patients and their prognosis.Cox regression model was applied to analyze the factors affecting the poor prognosis of TNBC patients.Results The level of miR-136-5p in cancer tissues(0.81±0.14)of TNBC patients was lower than that in adjacent tis-sues(1.02±0.21),and the difference was statistically significant,t=10.912,P<0.001;while the level of FAM83H(1.39±0.27)was higher than that in adjacent tissues(0.99±0.17),and the difference was statistically significant,t=16.442,P<0.001.There was a obvious negative correlation between miR-136-5p and FAM83H expression levels in cancer tissue of TNBC patients,r=-0.652,P<0.001.The level of miR-136-5p in cancer tissues(0.72±0.09)of TN-BC patients with lymph node metastasis was lower than that without lymph node metastasis(0.87±0.17),while the lev-el of FAM83H(1.51±0.38)was higher than that in patients without lymph node metastasis(1.31±0.20),and the difference was statistically significant,t values were 6.685,4.502,all P<0.05,respectively;the level of miR-136-5p in cancer tissues(0.71±0.11)of TNBC patients with vascular invasion was lower than that in patients without vascular in-vasion(0.83±0.15),while the level of FAM83H(1.54±0.33)was higher than that without vascular invasion(1.36±0.26),and the difference was statistically significant,t values were 4.087,3.240,all P<0.05,respectively.Five out of 172 TNBC patients were lost after 5 years of follow-up,and the incidence of poor prognosis in the remaining 167 patients was 24.55%.The incidence of poor prognosis in the miR-136-5p high expression group(15.79%)was lower than that in the low expression group(31.87%),and the difference was statistically significant,=6.252,P=0.012;the incidence of poor prognosis in the FAM83H high expression group(35.00%)was higher than that in the FAM83H low expression group(14.94%),and the difference was statistically significant,x2=11.522,P<0.001.Multivariate Cox regression a-nalysis showed that lymph node metastasis,vascular invasion,and FAM83H expression in cancer tissue were all risk fac-tors for poor prognosis in TNBC patients,while miR-136-5p expression in cancer tissue was a protective factor,all P<0.05.Conclusions The expression of miR-136-5p in cancer tissue of TNBC patients is down-regulated,while the expres-sion of FAM83H is up-regulated.The expression levels of miR-136-5p and FAM83H in cancer tissue are negatively corre-lated.Additionally,the expression of miR-136-5p and FAM83H in cancer tissue is closely related to the clinical patholog-ical characteristics and long-term prognosis of TNBC patients.
