[目的]探讨脓毒症大鼠肾小管上皮细胞中NGAL、IL-18的表达意义。[方法]采用盲肠结扎穿刺法建立脓毒症大鼠模型,检测脓毒症大鼠血清中肾小管上皮细胞中IL-18和NGAL的表达水平和铁离子浓度。铁离子处理后,检测肾小管上皮细胞中IL-18、NGAL水平、细胞焦亡水平。NGAL蛋白或IL-18抗体治疗后,对脓毒症大鼠的组织损伤情况进行评分。[结果]脓毒症大鼠肾小管上皮细胞中IL-18、NGAL的水平(0。09±0。02 vs 0。33±0。03,0。02± 0。01 vs 0。24±0。02,P<0。05)和铁离子浓度上升[(15。24±2。17)μmol/L vs(48。68±5。97)μmol/L,P<0。05]。铁离子处理后,肾上皮细胞中 IL-18、NGAL 的表达水平上升(0。10±0。02 vs 0。35±0。03,0。02±0。01 vs 0。24±0。02,P<0。05)、细胞焦亡水平上升(4。76±2。17vs38。11±7。65,P<0。05)。NGAL蛋白或IL-18抗体治疗后,脓毒症大鼠肾皮质(2。64±0。18vs 1。51±0。21,P<0。05)和外髓质损伤有所缓解(2。24±0。19 vs 1。52±0。22,P<0。05)。[结论]脓毒症诱导的大鼠肾小管上皮细胞NGAL、IL-18的表达水平较高。NGAL蛋白或者IL-18抗体治疗能够缓解脓毒症引起的急性肾损伤。
Expression significance of NGAL and IL-18 in renal tubular epithelial cells of sepsis rats
[Objective]To investigate the expression significance of NGAL and IL-18 in renal tubular epithelial cells of sepsis rats.[Method]Sepsis rat model was established by cecal ligation puncture method,and the expression levels of IL-18 and NGAL and the concentration of Fe3+in renal tubular epithelial cells in serum of sepsis rats were detected.The levels of IL-18,NGAL and pyrogen in renal tubular epithelial cells were detected after Fe3+ion treatment.Tissue damage in sepsis rats was scored after treatment with NGAL protein or IL-18 antibody.[Result]The levels of IL-18 and NGAL in renal tubular epi-thelial cells of sepsis rats(0.09±0.02 vs 0.33±0.03,0.02±0.01 vs 0.24±0.02,P<0.05)and Fe3+ion concentration in-creased[(15.24±2.17)μmol/L vs(48.68±5.97)μmol/L,P<0.05].After the Fe3+ion treatment,the expression levels of IL-18 and NGAL in renal epithelial cells increased(0.10±0.02 vs 0.35±0.03,0.02±0.01 vs 0.24±0.02,P<0.05),and the level of pyroptosis in renal epithelial cells increased(4.76±2.17 vs 38.11±7.65,P<0.05).After treatment with NGAL protein or IL-18 antibody,the damage of renal cortex(2.64±0.18 vs 1.51±0.21,P<0.05)and extramedullary cor-tex in sepsis rats was relieved(2.24±0.19 vs 1.52±0.22,P<0.05).[Conclusion]The expression levels of NGAL and IL-18 in renal tubular epithelial cells induced by sepsis were higher.At the same time,treatment with NGAL protein or IL-18 an-tibody alleviated acute kidney injury caused by sepsis.
