α-mating factor在毕赤酵母中引导蛋白分泌表达研究进展

毕赤酵母表达系统广泛用于外源蛋白分泌表达,如何提高外源蛋白的表达效率在毕赤酵母中的表达量是该系统的研究重点,分泌蛋白通过分泌途径的效率是限制外源蛋白表达的一个重要制约因素。α-mating factor信号序列常用于毕赤酵母中,酿酒酵母α-mating factor信号序列是毕赤酵母表达系统应用最早的一条信号序列,可引导大部分蛋白在毕赤酵母中的分泌表达,同时存在其他物种的α-mating factor信号序列能够在毕赤酵母中引导蛋白分泌表达。基于α-mating factor信号序列在毕赤酵母表达系统中的重要性,该文系统阐述了其在毕赤酵母中的研究进展,并描述了其结构和功能特点,提出了分析与思考,对α-mating factor信号序列的未来研究进行展望。
Research progress of protein secretion mediated by α-mating factor in Pichia pastoris
The Pichia pastoris expression system is widely used for the secretion and expression of heterologous proteins.How to improve the expression levels of heterologous proteins is the focus of the research in the system.The efficiency of secreted proteins through the secretory pathway is a limiting factor for heterologous protein production.The α-mating factor signal lead-er is commonly used in Pichia pastoris and the α-mating factor signal leader from Saccharomyces cerevisiae,which can mediate the secretion and expression of most proteins in P.pastoris and alpha-mating factor of other species guided protein expression in Pichia pastoris.Based on the important role of S.cerevisiae α-mating factor secretion signal in the production of recombinant proteins in P.pastoris expression system,in this paper,we systematically review its research progress on protein secretion in P.pastoris.The structural and functional characteristics of α-mating factor are described and the analysis are put forward.The fu-ture research of α-mating factor is prospected.
