Effect of imperatorin on rheumatoid arthritis in rats
[Objective]To explore the pharmacodynamic effect of europretin in the treatment of rheumatoid arthritis based on SUCNR1/IL-1β signaling pathway.[Method]100 clean-grade SD rats were randomly divided into normal control group,model control group,dexamethasone group(100.0 mg/kg),low and high dose imperatorin group,except for the normal control group,other groups were treated with bovine Ⅱ The rheumatoid arthritis model was established by induction of collagen type 2.Each drug group was given corresponding drug treatment,and the normal control group and model control group were given e-qual volumes of normal saline.After 4 weeks of treatment,the ankle joint inflammation index,the swelling value of the front paw,the levels of serum inflammatory factors IL-4,IL-8,and TNF-α,and the levels of SUCNR1,IL-1β mRNA and protein in the synovial tissue of the ankle joint were measured.[Result]Compared with the normal control group,the ankle joint inflam-mation index,front paw swelling value,serum IL-4,IL-8,TNF-α level,ankle joint synovium SUCNR1,IL-1β mRNA and protein expression in the model control group were increased high(P<0.05).Compared with the model control group,the an-kle joint inflammation index,forepaw swelling value,serum IL-4,IL-8,TNF-α levels,ankle synovium SUCNR1 levels in the low and high dose imperatorin group and the dexamethasone group,IL-1β mRNA and protein expression decreased(P<0.05).[Conclusion]Imperatorin has obvious pharmacodynamic effects on rheumatoid arthritis in rats,and its mechanism may be related to the inhibition of SUCNR1/IL-1β signaling pathway activation by imperatorin.
