摘要
[目的]运用杂交瘤技术制备二乙酰精胺(DAS)单克隆抗体,并对其进行鉴定及应用研究.[方法]通过小鼠制备DAS单克隆抗体,对抗体进行ELISA和POCT法进行鉴定.制备免疫比浊法试剂进行验证.[结果]获取特异性单克隆抗体,经鉴定制备出的DAS单克隆抗体纯度合格且稳定性好,制备了 DAS免疫比浊试剂盒.二乙酰精胺(DAS)在184例肠癌中的检出率为65.22％;CEA在170例肠癌中的检出率为40.00％;AFP在93例肠癌中的检出率为2.15％;CA199在166例肠癌中的检出率为17.47％;CA724在30例肠癌中的检出率为50％;CA125在87例肠癌中的检出率为14.94％.[结论]制备出的DAS单克隆抗体可作为诊断肠癌的标志物,结合其他标志物指标具有临床诊断价值.
Abstract
[Objective]To prepare diacetylspermine(DAS)monoclonal antibodies by hybridoma technique,and to character-ize them and study their applications.[Method]Diacetylspermine(DAS)immunogen was prepared and immunized in Balb/c healthy female mice,and the mouse spleen cells were fused with SP2/0 myeloma cells.The diacetylspermine(DAS)antibody was purified,specific antibody identification and clinical application.[Result]The specific monoclonal antibody was obtained,the purity of DAS monoclonal antibody was qualified and the stability was good,and the DAS immunoturbidimetric kit was made.The detection rate of diacetylspermine(DAS)in 184 cases of colorectal cancer was 65.22％.The detection rate of CEA in 170 cases of colorectal cancer was 40.00％.The detection rate of AFP in 93 cases of colorectal cancer was 2.15％.The de-tection rate of CA199 in 166 cases of colorectal cancer was 17.47％.The detection rate of CA724 in 30 cases of colorectal cancer was 50％.The detection rate of CA125 in 87 cases of colorectal cancer was 14.94％.[Conclusion]Diacetylspermine(DAS)monoclonal antibody with good specificity and stability was successfully prepared and can be used for the early clinical diagnosis of intestinal,hepatocellular and gastric cancers.
