[目的]从轮钟花的根中提取出蜘蛛果多糖(Spider fruit polysaccharide)并研究其对RAW264。7细胞的免疫调控作用。[方法]通过超声提取法提取蜘蛛果多糖,CCK-8法检测(脂多糖)LPS对RAW264。7细胞增殖的情况,硝酸还原酶法测定蜘蛛果多糖对RAW264。7细胞代谢NO的水平,ELISA法测定蜘蛛果多糖对RAW264。7细胞分泌TNF-α、1L-6水平的情况。[结果]蜘蛛果多糖在25 μg/mL~400 μg/mL浓度范围内能促进RAW264。7细胞的增殖;与空白对照组相比,蜘蛛果多糖能显著增加RAW264。7细胞的NO、IL-6及TNF-α分泌量,与LPS模型组相比,则能显著降低RAW264。7细胞的NO、TL-6及TNF-α分泌量。[结论]蜘蛛果多糖可以激活巨噬细胞,调节机体免疫力。
Immunomodulatory effect of Campanumoeae lancifoliorum Radix polysaccharides on RAW264.7 cells
[Objective]To study the immunoregulatory activity of Campanumoeae lancifoliorum Radix polysaccharides on RAW264.7 cells.[Method]Campanumoeae lancifoliorum Radix polysaccharides were extracted by ultrasonic extraction.The proliferation of RAW264.7 cells was detected by CCK-8 method.The effect of Campanumoeae Lancifoliorum Radix polysac-charides on the secretion of NO in RAW264.7 cells was determined by nitrate reductase method.The effect of Campanumoeae lancifoliorum Radix polysaccharides on the secretion of TNF-α and IL-6 in RAW264.7 cells was determined by ELISA method.[Result]Campanumoeae lancifoliorum Radix polysaccharides could promote the proliferation of RAW264.7 cells in the concentration range of 25 μg/mL-400 μg/mL,and significantly reduce the secretion of NO,IL-6 and TNF-α by RAW264.7 cells induced by LPS.[Conclusion]Campanumoeae lancifoliorum Radix polysaccharides has a certain regulatory effect on the immune system.
