Isolation and identification of the pathogen of honeysuckle root rot and analysis of its biological characteristics
[Objective]In order to clarify the causes of the phenomena such as leaf loss,yellowing,browning and drying,root browning,and attachment of white mycelium on the surface of the stem base,some plants in honeysuckle fields in Julu County,Hebei Province,China,in recent years.[Method]The diseased strain was isolated by tissue isolation method,its pathogenicity was verified using Koch's law,and the strain species was identified by morphology and ITS sequences,followed by biological characterization of the strain.[Result]A pathogenic strain B22 was isolated,and the homology with Fusarium equiseti was more than 99%,which was identified as Fusarium equiseti.The results of biological characterization showed that the OMA medium was the most suitable for the growth of its mycelium,the optimal growth temperature was 28 ℃,the optimal pH value was 7,the effect of light on it was small,and the lethal temperature was 61 ℃.[Conclusion]The pathogenic bacterium,namely Fusarium equiseti,was isolated and identified from the honeysuckle disease,and its biological characteristics were analyzed,This strain grew fastest at 28 ℃ in OMA culture medium at pH 7,and was less affected by light,but was lethal at 61 ℃ for 10 min.This study can provide a relatively objective basic theoretical basis for the diagnosis,prevention and control of honeysuckle homoge-neous diseases.
