Effect of extract of Radix Adenophora on lung inflammation in rats with radiation lung injury
[Objective]To explore the effect of extract of Radix Adenophora on lung inflammation in rats with radiation lung in-jury.[Method]The rats were randomly divided into 6 groups:Sham operation group,model group,positive control group and low-dose,medium-dose and high-dose groups(18.75 mg/kg,37.50 mg/kg,75.00 mg/kg)of extract of Parapsychia palma-ta,with 12 animals in each group,were irradiated by linear accelerator at a single time of 15 Gy to establish radiation-induced lung injury animal models,and were given continuous administration for 28 days after irradiation,and killed after modeling.The pathological changes of the lung tissues were evaluated by HE and Masson staining,the serum inflammatory factors were detec-ted by ELISA,the mRNA expression levels of inflammatory factors and key proteins in the regulation of inflammatory infiltration in the lung tissues were detected by real-time fluorescent quantitative PCR,and the expression levels of proteins related to sig-naling pathways in the lung tissues were detected by Western Blot.[Result]HE staining showed that compared with sham oper-ation group,alveolar inflammation was significantly increased in model group(P<0.05),and inflammatory cell infiltration was decreased in each dose group(P<0.05).Compared with the sham operation group,the levels of TGF-β1,TNF-α,IL-1βand IL-6 in serum of rats in the other groups were as follows:The expression levels of TGF-β 1,p-SMad2/3 protein and IL-1β,IL-6,TNF-α,TGF-β1,α-SMA and COL1 A1 mRNA in lung tissues were significantly increased(P<0.05),while the expression levels of intervention groups were significantly decreased compared with model group(P<0.05).There was a dose-response relationship(P<0.05).[Conclusion]The extract of Radix Adenophora has protective effect on acute lung in-jury caused by radiation,and its mechanism may be closely related to reducing inflammatory factors and changing the expression of inflammatory regulatory proteins.
