Analysis of genes of peripheral blood mononuclear cells in allergic rhinitis and treatment with traditional Chinese medicine
[Objective]To analyze the key genes and pathways in peripheral blood single nucleated cells of allergic rhinitis based on bioinformatics methods,and to carry out the association analysis of therapeutic Chinese medicines.[Method]We ana-lyzed the differential genes in peripheral blood single nucleated cells of patients with allergic rhinitis on the GSE50223 microar-ray,used the differential genes to enrich the functions,pathways and mechanisms,screened out the key targets and associated with related therapeutic Chinese medicines,and analyzed the properties and efficacy of therapeutic Chinese medicines that inter-vene in the key targets.[Result]GSE50223 microarray analysis yielded 684 differential target genes.The regulation of AR by differential genes was mainly related to TLRs signaling pathway,PPAR signaling pathway,IL-17 signaling pathway,etc.Ten key targets were obtained,which were CCNA2,TOP2A,CCNB2,CDC20,CDCA8,KIF11,KIF20A,BUB1,CCNB1,and KIF2C,respectively.153 types of traditional Chinese medicines were obtained.The highest frequency of association was between Kuxin-gren(armeniacae semen amarum)and Hongqi(hedysari radix).The associated Chinese medicines were mainly cold-natured and warm-natured,mostly sweet and pungent,mostly entered the liver meridian of foot queyin and lung meridian of hand taiy-in,and were mostly heat-clearing and deficiency-tonifying medicines.[Conclusion]Based on the biosignature analysis,10 differentially expressed key genes in AR were identified,which were involved in the pathway mechanisms of the immune sys-tem,cell cycle,and hemostasis,and potential drugs such as Kuxingren(armeniacae semen amarum),Hongqi(hedysari radix),Aiye(artemisiae argyi folium),and Baiguo(ginkgo semen)were screened for possible prevention and treatment of allergic rhi-nitis,which provided data support for the molecular pathogenesis of AR and the use of traditional Chinese medicines.
