Predictive value of Runx2,RANKL and GPR48 in the onset of hormone-induced osteonecrosis of the femoral head
[Objective]To analyze the expression of Runx2,RANKL and GPR48 in steroid induced osteonecrosis of the femoral head in rats and their predictive value.[Method]20 rats were randomly divided into model group and control.In model group,glucocorticoid was injected intramuscularly to establish steroid induced femoral head necrosis model.After successful modeling,HE staining was performed in the two groups,and the levels of Runx2,RANKL and GPR48 in the lesion tissue were detected by Western Blotting and RT-PCR.The relationship between Runx2,RANKL and GPR48 and steroid induced femoral head necro-sis was analyzed by logistic multiple regression model.[Result]In the model group,tissue cell atrophy,cell structure disorder,cell aggregation and pyknosis were observed,and there were obvious empty bone lacunae;the maximum diameter of adipocytes and empty bone lacunae rate in the model group were obvious higher than control group(46.38 μm±7.49 μm vs 23.81 μm±6.18 μm;20.93%±2.94%vs 7.67%±1.04%).And the trabecular area was lower than that control group(28.74%±4.53%vs 60.95%±5.07%),and the difference was statistically obvious(P<0.05);the RANKL protein and mRNA levels in the model group were obvious higher than control group(5.92%±0.14%vs 1.29%±0.07%;2.89%±0.12%vs 0.21%±0.05%).The levels of Runx2,GPR48 were obvious lower than control group(0.91%±0.11%vs 2.83%±0.21%;1.34%±0.12%vs 2.83%±0.09%).The levels of Runx2,RANKL and GPR48 in steroid induced osteonecrosis of the femoral head were independent risk factors for the pathogenesis of steroid induced osteonecrosis of the femoral head(P<0.05).[Conclu-sion]Runx2 and GPR48 were highly expressed and RANKL was low expressed in rats with steroid induced osteonecrosis of the femoral head.Low Runx2,high RANKL and low GPR48 levels were independent risk factors for the pathogenesis of steroid in-duced osteonecrosis of the femoral head in rats.
