[目的]探究石斛多糖对吞咽障碍大鼠舌下神经核内神经元活性的影响。[方法]将大鼠随机分为3组:对照组、模型组和治疗组,每组各10只大鼠。治疗组的大鼠使用石斛多糖(0。33 g/kg)处理1 w,每日1次。对照组和模型组的大鼠使用磷酸盐缓冲液处理。通过尼氏染色分析神经元数量;MTT试验检测神经细胞生长能力;通过流式细胞术分析神经细胞凋亡率;通过蛋白免疫印迹分析Bax和Bel-2蛋白的表达水平。[结果]与模型组比较,治疗组舌下神经核尼氏小体的数量显著增加(51。96±6。05 vs 23。31±2。12,P<0。05);舌下神经核内神经细胞增殖能力显著增加(0。31±0。02 vs 0。46±0。02,P<0。05);舌下神经核内神经细胞凋亡率显著降低(17。23%±1。29%vs 35。28%±1。32%,P<0。05);舌下神经核内神经细胞Bax/Bcl-2蛋白表达比值显著降低(0。46±0。06 vs 0。92±0。02;0。43±0。05 vs 0。22±0。02,P<0。05)。[结论]石斛多糖能够提高吞咽障碍大鼠舌下神经核内神经元活性,对于吞咽障碍可能具有一定疗效。
Effect of dendrobium polysaccharides on neuronal activity in hypoglossal nucleus of rats with dysphagia
[Objective]To investigate the effects of dendrobium polysaccharides on the activity of neurons in hypoglossal nu-cleus in rats with dysphagia.[Method]Rats were randomly divided into 3 groups:control group,model group and treatment group,with 10 rats in each group.The rats in the treatment group were treated with dendrobium polysaccharide(0.33 g/kg)once a day for 1 week.The rats in the control and model groups were treated with phosphate buffer solution.The number of neu-rons was analyzed by Nissl staining.MTT assay was used to detect the growth ability of nerve cells.The apoptosis rate of nerve cells was analyzed by flow cytometry.The expression levels of Bax and Bcl-2 proteins were analyzed by Western Blot.[Re-sult]Compared with the model group,the number of Nissl bodies in the hypoglossal nerve nucleus was significantly increased in the treatment group(51.96±6.05 vs 23.31±2.12,P<0.05).The proliferation of nerve cells in hypoglossal nucleus was sig-nificantly increased(0.31±0.02 vs 0.46±0.02,P<0.05).The apoptosis rate of nerve cells in hypoglossal nucleus was sig-nificantly decreased(17.23%±1.29%vs 35.28%±1.32%,P<0.05).The ratio of Bax/Bcl-2 protein expression in hypo-glossal nucleus was significantly decreased(0.46±0.06 vs 0.92±0.02;0.43±0.05 vs 0.22±0.02,P<0.05).[Conclu-sion]Dendrobium polysaccharides can improve the activity of neurons in hypoglossal nucleus in dysphagia rats,and may have a certain effect on dysphagia.
