酿酒用糯高粱生长过程需要大量的磷元素,缺磷会造成糯高粱的减产,但是化肥的大量施用会造成土壤肥力下降,为开发促进糯高粱生长的溶磷菌剂,使用溶磷圈法从糯高粱根内筛选具有溶磷功能的菌株,对获得的菌株进行菌种鉴定,并将该菌株制成菌悬液接种到糯高粱根部,测定糯高粱的生长指标,评估该菌株对糯高粱的促生长性能.从糯高粱根中筛选得到了6株溶磷菌,其中溶磷圈直径与菌落直径比值最大的是菌株GP4-1,为2.2.基于菌株16S rDNA序列的系统发育分析,GP4-1属于伯克霍尔德菌(Burkholderia sp.).将GP4-1菌悬液接种到高粱幼苗根部30 d后,高粱幼苗的株高增加了4.46%,鲜重增加了19.69%,干重增加了100%,植株全磷含量增加了2.65%.糯高粱根内生菌GP4-1通过促进糯高粱植株对磷元素的吸收促进高粱的生长,为针对糯高粱的溶磷菌剂的开发和应用奠定了基础.
Isolation and Screening of Phosphate Solubilizing Bacteria and its Growth-promoting Effect on Glutinous Sorghum for Brewing
The growth process of glutinous sorghum for brewing requires a large amount of phosphorus element,and the lack of phosphorus would result in the yield reduction,but the long time of fertilizer use would cause the decline of soil fertility.In order to develop the phosphorus soluble bacteria agent to promote glutinous sorghum growth,this study used the phosphorus soluble ring method to screen the strains with phosphorus soluble function from glutinous sorghum roots,and the strains obtained were identified.The suspension was inoculated into the root of glutinous sorghum seedlings,the glutinous sorghum growth traits were determined,and the growth promotion performance of the strain on glutinous sorghum was evaluated.In this study,six strains of phosphorus solubilizing bacteria were selected from glutinous sorghum roots,isolate GP4-1 had the largest ratio of phosphorus solubilizing circle diameter to colony diameter,which was 2.2.Based on phylogenetic analysis of 16S rDNA sequences,GP4-1 belonged to Burkholderia sp.GP4-1.After 30 days of inoculation with GP4-1 bacterial suspension,plant height,fresh weight,dry weight and the total phosphorus content of sorghum seedlings increased by 4.46%,19.69%,100%and 2.65%,respectively.The root endophyte GP4-1 promoted glutinous sorghum growth by promoting phosphorus absorption.This study laid a foundation for the development and application of phosphorus solubilizing bacteria for glutinous sorghum.
