Bipolaris AustraliensisHD-1产胆红素氧化酶的培养基组份及发酵条件优化

为了更大限度地获得徽生物胞外分泌物产量,利用部分因子,最陡爬坡及BoxBenken(BB)试验及其设计对Bipolaris australiensis HD-1胞外产胆红素氧化酶(bilirubin oxidase,BOD)的培养基组份及发酵条件进行了优化,结果表明,当培养基质中初始组成含量(g/L)分别为麦芽糖5,硫酸铜0.32,大豆蛋白胨50;发酵温度为29.8℃,pH6.8,摇床转速为150rpm,在有氧条件下培养6d可获得2.18 U/mL的酶产量,其产酶量比优化前提高了近3倍.
Optimization Culture Medium Composition and Fermentation Control of Bilirubin Oxidase Producing from Bipolaris Australiensis HD-1 by a Series of Statistics Methods
In order to obtained more production of extracellar secretion of microorganism,a series of statistics methods containing 2-level factorial design,steepest ascent design and BoxBenken design of response surface method respectively were applied with optimization medium composition and fermentation control of bilirubin oxidase from Bipolaris australiensis HD-1,of which results shown that medium composition containing(g/L):maltose 5,CuSO45H2O 0.32 and soy peptone 50 at temperature of 29.8℃,pH6.8,aerobics stir 150 rpm for 6 d the enzyme activity be up to 2.18 U/mL,of which about threefold more than one of non-optimization.
