An Optimization Algorithm for the Kinetics Mechanism Discrimination of The Double Substrates Enzymatic Reaction
A common used method to research the kinetics mechanism of double substrates enzymatic reaction is double-reciprocal plots.In this paper,an optimization algorithm based on the last squares fitting principle is proposed to solve the disadvantages in double-reciprocal plots method.In the optimization algorithm,the constant coefficients of the double-reciprocal kinetics equations for both sequential mechanism and ping-pong mechanism are calculated by the last squares fitting.The kinetics mechanism of the enzymatic reaction is distinguished through the comparison of residual sum of squares for the two curve-fitting functions.An example is specified to prove that the optimization algorithm is concise and reliable,and is more accurate in the double substrates enzymatic reaction kinetics mechanism discrimination.
